How to Reconstitute Retatrutide: Step-by-Step BAC Water & Syringe Math Guide
10mg Vial + 2mL BAC Water = 5.0 mg/mL (50 mcg per 1 Syringe Unit on U-100)
Standard Diluent: 0.9% Benzyl Alcohol Bacteriostatic Water (USP)
Required Laboratory Equipment
- Retatrutide Lyophilized Powder Vial (5mg, 10mg, or 15mg)
- Bacteriostatic Water (0.9% Benzyl Alcohol) — Multi-dose sterile preservative diluent
- 70% Isopropyl Alcohol Prep Pads
- 3mL Luer-Lock Syringe with 21G - 25G Needle (for reconstitution)
- U-100 31G Insulin Syringes (for research dosing)
Step-by-Step Reconstitution Protocol
- Disinfect the rubber stoppers of both the Bacteriostatic Water vial and the Retatrutide lyophilized peptide vial with fresh 70% isopropyl alcohol swabs. Allow to air dry for 30 seconds.
- Draw exactly 2.0 mL of air into a 3mL mixing syringe. Inject the air into the BAC water vial and withdraw 2.0 mL of bacteriostatic water.
- Insert the mixing needle into the Retatrutide vial at a 45-degree angle against the inner glass wall. Release the water slowly down the side of the glass. NEVER squirt liquid directly onto the lyophilized powder cake.
- Allow the vacuum inside the vial to pull the plunger naturally if present. Remove the needle once liquid transfer is complete.
- Gently roll the vial between your palms for 60-90 seconds. DO NOT shake or vortex the vial, as peptide chains are delicate and can shear.
- Inspect the reconstituted solution: it should be crystal clear with zero floating particulate matter.
- Store immediately in the refrigerator between 2°C - 8°C (36°F - 46°F). Protect from direct light.
Syringe Draw Volume by Target Dose
| Target Dose (mcg) | U-100 Syringe Draw | U-50 Syringe Draw |
|---|---|---|
| 1000 mcg | 20 Units (0.2 mL) | 10 Units |
| 2000 mcg | 40 Units (0.4 mL) | 20 Units |
| 4000 mcg | 80 Units (0.8 mL) | 40 Units |
Safety & Sterility Guidelines (USP <797>)
- Never reuse needles or mixing syringes.
- Always check that the BAC water is clear and not past its 28-day puncture limit.
- Do not freeze reconstituted Retatrutide; freeze-thaw cycles destroy peptide quaternary structure.
